Ample nitrogen (N) is required for potato production, but its use efficiency is low. N supply strongly interacts with maturity type of the cultivar grown. We assessed whether variation among 189 cultivars grown with 75 or 185 kg available N/ha in 2 years would allow detecting quantitative trait loci (QTLs) for relevant traits. Using phenotypic data, we estimated various traits and carried out a genome-wide association study (GWAS) with kinship correction. Twenty-four traits and 10,747 markers based on single-nucleotide polymorphisms from a 20K Infinium array for 169 cultivars were combined in the analysis. N level affected most traits and their interrelations and influenced the detection of marker-trait associations; some were N-dependent, others were detected at both N levels. Ninety percent of the latter accumulated on a hotspot on Chromosome 5. Chromosomes 2 and 4 also contained regions with multiple associations. After correcting for maturity, the number of QTLs detected was much lower, especially of those common to both N levels; however, interestingly, the region on Chromosome 2 accumulated several QTLs. There is scope for marker-assisted selection for maturity, with the main purpose of improving characteristics within a narrow range of maturity types, in order to break the strong links between maturity type and traits like N use efficiency.

How the growing world population can feed itself is a crucial, multi-dimensional problem that goes beyond sustainable development. Crop production will be affected by many changes in its climatic, agronomic, economic, and societal contexts. Therefore, breeders are challenged to produce cultivars that strengthen both ecological and societal resilience by striving for six international sustainability targets: food security, safety and quality; food and seed sovereignty; social justice; agrobiodiversity; ecosystem services; and climate robustness. Against this background, we review the state of the art in plant breeding by distinguishing four paradigmatic orientations that currently co-exist: community-based breeding, ecosystem-based breeding, trait-based breeding, and corporate-based breeding, analyzing differences among these orientations. Our main findings are: (1) all four orientations have significant value but none alone will achieve all six sustainability targets; (2) therefore, an overarching approach is needed: "systems-based breeding," an orientation with the potential to synergize the strengths of the ways of thinking in the current paradigmatic orientations; (3) achieving that requires specific knowledge development and integration, a multitude of suitable breeding strategies and tools, and entrepreneurship, but also a change in attitude based on corporate responsibility, circular economy and true-cost accounting, and fair and green policies. We conclude that systems-based breeding can create strong interactions between all system components. While seeds are part of the common good and the basis of agrobiodiversity, a diversity in breeding approaches, based on different entrepreneurial approaches, can also be considered part of the required agrobiodiversity. To enable systems-based breeding to play a major role in creating sustainable agriculture, a shared sense of urgency is needed to realize the required changes in breeding approaches, institutions, regulations and protocols. Based on this concept of systems-based breeding, there are opportunities for breeders to play an active role in the development of an ecologically and societally resilient, sustainable agriculture.

Increasing demand for nutritious, safe, and healthy food because of a growing population, and the pledge to maintain biodiversity and other resources, pose a major challenge to agriculture that is already threatened by a changing climate. Diverse and healthy diets, largely based on plant-derived food, may reduce diet-related illnesses. Investments in plant sciences will be necessary to design diverse cropping systems balancing productivity, sustainability, and nutritional quality. Cultivar diversity and nutritional quality are crucial. We call for better cooperation between food and medical scientists, food sector industries, breeders, and farmers to develop diversified and nutritious cultivars that reduce soil degradation and dependence on external inputs, such as fertilizers and pesticides, and to increase adaptation to climate change and resistance to emerging pests.

Robustness in lettuce, defined as the ability to produce stable yields across a wide range of environments, may be associated with below-ground traits such as water and nitrate capture. In lettuce, research on the role of root traits in resource acquisition has been rather limited. Exploring genetic variation for such traits and shoot performance in lettuce across environments can contribute to breeding for robustness. A population of 142 lettuce cultivars was evaluated during two seasons (spring and summer) in two different locations under organic cropping conditions, and water and nitrate capture below-ground and accumulation in the shoots were assessed at two sampling dates. Resource capture in each soil layer was measured using a volumetric method based on fresh and dry weight difference in the soil for soil moisture, and using an ion-specific electrode for nitrate. We used these results to carry out an association mapping study based on 1170 single nucleotide polymorphism markers. We demonstrated that our indirect, high-throughput phenotyping methodology was reliable and capable of quantifying genetic variation in resource capture. QTLs for below-ground traits were not detected at early sampling. Significant marker-trait associations were detected across trials for below-ground and shoot traits, in number and position varying with trial, highlighting the importance of the growing environment on the expression of the traits measured. The difficulty of identifying general patterns in the expression of the QTLs for below-ground traits across different environments calls for a more in-depth analysis of the physiological mechanisms at root level allowing sustained shoot growth.

Organic agriculture requires cultivars that can adapt to organic crop management systems without the use of synthetic pesticides as well as genotypes with improved nutritional value. The aim of this study encompassing 16 experiments was to compare 23 broccoli cultivars for the content of phytochemicals associated with health promotion grown under organic and conventional management in spring and fall plantings in two broccoli growing regions in the US (Oregon and Maine). The phytochemicals quantified included: glucosinolates (glucoraphanin, glucobrassicin, neoglucobrassin), tocopherols (δ-, γ-, α-tocopherol) and carotenoids (lutein, zeaxanthin, ß-carotene). For glucoraphanin (17.5%) and lutein (13%), genotype was the major source of total variation; for glucobrassicin, region (36%) and the interaction of location and season (27.5%); and for neoglucobrassicin, both genotype (36.8%) and its interactions (34.4%) with season were important. For δ- and γ-tocopherols, season played the largest role in the total variation followed by location and genotype; for total carotenoids, genotype (8.41-13.03%) was the largest source of variation and its interactions with location and season. Overall, phytochemicals were not significantly influenced by management system. We observed that the cultivars with the highest concentrations of glucoraphanin had the lowest for glucobrassicin and neoglucobrassicin. The genotypes with high concentrations of glucobrassicin and neoglucobrassicin were the same cultivars and were early maturing F1 hybrids. Cultivars highest in tocopherols and carotenoids were open pollinated or early maturing F1 hybrids. We identified distinct locations and seasons where phytochemical performance was higher for each compound. Correlations among horticulture traits and phytochemicals demonstrated that glucoraphanin was negatively correlated with the carotenoids and the carotenoids were correlated with one another. Little or no association between phytochemical concentration and date of cultivar release was observed, suggesting that modern breeding has not negatively influenced the level of tested compounds. We found no significant differences among cultivars from different seed companies.
